چکیده

Objective: Overexpression of SLUG has been implicated in promoting migration and invasion of many cancer cells, including anaplastic thyroid cancer (ATC) cells. Thus, targeted down-regulation of SLUG expression in ATC cells and determination of its implications would provide new treatment approaches for disease management. Methods: Small interfering RNA (siRNA) targeting SLUG (SLUG siRNA) was used to develop clonal derivatives of the ATC SW1736 cells. SLUG cDNA transfection was used to restore the SLUG expression in the SLUG siRNA transfected SW1736 cells. E-cadherin siRNA was used to inhibit E-cadherin expression in the SLUG siRNA transfected SW1736 cells. The effect of SLUG and E-cadherin expression on migration and invasion of SW1736 cells in vitro was detected. Further, the effect of targeting SLUG on lung metastasis in an mouse SW1736 cell tumor model was detected. Results: Targeting SLUG expression by SLUG siRNA transfection in SW1736 cells showed a 75% decrease in migration and a 85% decrease in invasion in vitro. Targeting E-cadherin by E-cadherin siRNA transfection or SLUG overexpression by SLUG cDNA transfection restored the invasion and migration ability of SW1736 cells. In vivo analysis indicated a 80% decrease in the number of mice bearing macroscopic lung metastases. Analysis of SLUG signaling pathways in the clonal derivatives showed a decrease in SLUG expression and increase in E-cadherin expression in the SLUG siRNA transfected tumor. Conclusions: Therapies targeting SLUG signaling pathway may be more effective in preventing organ metastasis.

Effect of Slug-Mediated Down-Regulation of E-Cadherin on Invasiveness and Metastasis of Anaplastic Thyroid Cancer Cells

BACKGROUND Slug has been found to promote migration and invasion of many cancer cells, including anaplastic thyroid cancer (ATC). Thus, targeting Slug expression could provide new approaches for the treatment of patients with ATC. MATERIAL AND METHODS Small interfering RNA (siRNA) targeting Slug (Slug siRNA) was used to construct clonal derivatives in the metastatic ATC SW1736 cells. Slug cDNA ...

Slug promotes survival during metastasis through suppression of Puma-mediated apoptosis.

Tumor cells must overcome apoptosis to survive throughout metastatic dissemination and distal organ colonization. Here, we show in the Polyoma Middle T mammary tumor model that N-cadherin (Cdh2) expression causes Slug (Snai2) upregulation, which in turn promotes carcinoma cell survival. Slug was dramatically upregulated in metastases relative to primary tumors. Consistent with a role in metasta...

Autoregulation of E-cadherin expression by cadherin-cadherin interactions: the roles of β-catenin signaling, slug and MAPK

ranscriptional repression of E-cadherin , characteristic of epithelial to mesenchymal transition, is often found also during tumor cell invasion. At metastases, migratory fibroblasts sometimes revert to an epithelial phenotype, by a process involving regulation of the E-cadherin– -catenin complex. We investigated the molecular basis of this regulation, using human colon cancer cells with aberra...

Silencing of rhomboid domain containing 1 to inhibit the metastasis of human breast cancer cells in vitro

Objective(s): A growing body of evidence indicates that rhomboid domain containing 1 (RHBDD1) plays an important role in a variety of physiological and pathological processes, including tumorigenesis. We aimed to determine the function of RHBDD1 in breast cancer cells. Materials and Methods: In this study, we used the Oncomine™ database to determine the expression patterns of RHBDD1 in normal a...

Silibinin upregulates E-cadherin expression in MKN-45 human gastric cancer cells

Background and objectives:  Gastric cancer is currently known as one of the most important causes of cancer-driven death all over the world. In patients with gastric cancer, a significant proportion of death occurs due to metastasis. On the other hand, down modulated E-cadherin level has been reported as an important contributor to tumor cell invasion and metastasis. In this re...
